What are internal controls and how are they used in the data analysis?

Olink Explore contains three types of internal controls spiked into our assay that monitor every step of the procedure.

  1. Incubation Control is PEA assay for non-human proteins which monitor potential variation across all steps, and used in data QC.
  2. Extension Control is an antibody molecule with both DNA-oligos attached and therefore always in proximity. This controls for the extension, library preparation and sequencing steps and is also used for data normalization purposes.
  3. Amplification Control is a synthetic double-stranded amplicon which controls the library preparation and sequencing step of the protocol and used in data QC.
