Neprilysin (also known as membrane metallo-endopeptidase, neutral endopeptidase, or CD10), is a zinc-dependent metalloprotease that cleaves peptides at the amino side of hydrophobic residues. MME expression is considered as a marker for common lymphoid progenitor cells and its presence or absence is diagnostically useful for specific types of leukemias and lymphomas (e.g. is expressed in angioimmunoblastic T cell lymphoma and Burkitt lymphoma, but is normally absent in acute myeloid leukemia and mantle cell lymphoma). Neprilysin also has in important role in degrading the amyloid beta peptide whose abnormal misfolding and aggregation in neural tissue has been implicated as a cause of Alzheimer's disease. Both the declining expression of MME with age, as well as experimental evidence from knockout mice provide evidence that this protein may play a key role in Alzheimer's disease and could be a potential therapeutic target.
Recommended sample types are human EDTA plasma and serum. A range of additional sample types are compatible with the technology (PEA), including citrate plasma, heparin plasma, cerebrospinal fluid, (CSF), tissue and cell lysates, fine needle biopsis, microdialysis fluid, cell culture media, dried blood spots, synovial fluid, saliva, plaque extract and microvesicles. Please note that protein expression levels are expected to vary in different sample types. Certain assays are differentially affected by interfering substances such as hemolysate. NOTE: The calibrator curve below shows the performance of the assay with the estimated sensitivity and dynamic range parameters indicated. These curves are generated during the assay validation process using recombinant antigens. Please note that when analyzing biological samples the data generated will be given in the form of relative quantification (NPX values) and cannot be converted to absolute protein concentrations.
